The minimal photounit of the photosynthetic membranes of the purple non-sulfur bacterium Rhodospirillum rubrum, comprising the reaction center and the light-harvesting complex has been purified and crystd. in two dimensions in the presence of added phospholipids, and subsequently visualized by electron microscopy after neg.-staining. The position of the reaction centers within the light-harvesting ring has been detd. at low resoln. by the application of a new anal. for rotationally disordered identical unit

The invention concerns a method for screening compound interaction with transmembrane receptors involved in cellular signal transduction, preferably with G protein coupled receptors, by adding the compound to be analyzed into a medium comprising native inv ...

We have measured fluorescence resonance energy transfer (FRET) between a fluorescent antagonist, bound to the purified detergent-solubilized serotonin type 3 receptor, and a lipophilic acceptor probe partitioned into the micelle surrounding the detergent-solubilized receptor. The exptl. obsd. FRET efficiency was evaluated on the basis of the characteristic dimensions of the receptor-micelle complex and the av. no. of acceptor mols. in such micelles. The binding site was detd. to be 5.4 +- 0.9 nm above the c

The secondary structure of porin, maltoporin, and OmpA protein reconstituted in lipid membranes was detd. by Raman spectroscopy. The 3 proteins have similar structures consisting of 50-60% b-strand, .apprx.20% b-turn, and

5 HT receptors expressed throughout the human body are targets for established therapeutics and various drugs in development. Their diversity of structure and function reflects the important role 5-HT receptors play in physiologic and pathophysiological pr ...

A new strategy for the reversible formation of supported lipid bilayers is introduced. It uses an extension of immobilized metal ion affinity chromatog. technol., which depends on the reversible formation of complexes between metal ion binding chelator groups and oligohistidines. Lipid layers contg. hexahistidine-derivatized amphiphiles (His-lipids) are anchored to a self-assembled monolayer (SAM) contg. synthetic chelator thioalkanes (CTA). The control over the d. of anchor sites on the substrate and over

Neurotransmitter-gated ion channels of the Cys-loop receptor family mediate fast neurotransmission throughout the nervous system. The molecular processes of neurotransmitter binding, subsequent opening of the ion channel and ion permeation remain poorly un ...

Transmembrane protein 16 F (TMEM16F) is a Ca2+-activated homodimer which functions as an ion channel and a phospholipid scramblase. Despite the availability of several TMEM16F cryogenic electron microscopy (cryo-EM) structures, the mechanism of activation ...

The specific recognition reaction between cholera toxin (ChTo) and the ganglioside GM1 was investigated in situ on planar supported lipid layers using simultaneous surface plasmon resonance (SPR) and electrochem. impedance measurements. The lipid monolayers contg. different amts. of GM1 were formed on alkanethiol-coated gold surfaces by a new lipid/detergent diln. technique. The formation of the layers was investigated using both SPR and impedance measurements. While the optical properties of the different

The changes in bilayer structure induced by surface charges in the case of an ionizable lipid were studied by x-ray diffraction, Raman spectroscopy, and film-balance measurements. With increasing surface charge in the ordered phase, the x-ray results show a decrease in bilayer thickness, whereas the hydrocarbon chain packing stays essentially const.; Ramam data show that the internal chain ordering changes, and monolayer studies show a lateral expansion of the bilayer. These results are interpreted in terms

Polarized IR spectra of melittin (I) incorporated into macroscopically oriented lipid membranes are reported. From the linear dichroism of the amide I and amide II vibrational bands, the spatial orientation of I helices was detd. as being preferentially parallel to the membrane normal under exptl. condition of low water content and an ordered lipid phase. The data are in accord with an orientation of membrane-incorporated I whereby the hydrophobic part of the polypeptide either spans the bilayer in the form

The ability of organisms, or individual cells, to react to external chemical signals, which are detected and transduced by cell-surface receptors, is crucial for their survival. These receptors are the targets of the majority of clinically used medicines. Combinatorial genetics can provide almost unlimited numbers of mutant receptor proteins and combinatorial chemistry can produce large libraries of potential therapeutic compounds that act on these membrane receptors. What is missing for the fundamental und

The invention relates to a method and a device for sequencing nucleic acid mols. on a perforated membrane. The inventive method and device can be esp. used in multiplex sequencing. The method involves prepg. an oligonucleotide that is labeled to satn. with base-specific fluorescent dyes that is then immobilized on a filter. The labeled sequence is then processively degraded with an exonuclease with the released bases being detected fluorometrically. [on SciFinder (R)]

A review, with .apprx.75 refs., on (1) structural models of membrane proteins derived from Raman spectroscopy and prediction methods, (2) dynamics of model peptides, (3) proline in transmembrane helixes, and other topics. [on SciFinder (R)]

Putative transmembrane helixes of membrane proteins in general and channel proteins in particular often contain proline residues which may induce a bend into an otherwise regular helical structure. Here, it is shown by fluorescence-energy-transfer measurements and mol.-dynamics calcns. that, in the case of synthetic bilayer-spanning helical polypeptides, a proline-induced bend in a helix acts as a flexible element mediating rigid body motions of the helical segments. Most important, such structural fluctuat

Highly efficient capillary electrochromatog. sepns. of cardiac glycosides and other steroids are presented. Employing butyl-derivatized silica particles as stationary phase resulted in a nearly three times faster electroosmotic flow (EOF) compared to capillary electrochromatog. (CEC) with octadecyl silica particles. On-column focusing with a preconcn. factor of 180 was performed and sepn. efficiencies of up to 240 000 plates per m were obtained. Using label-free std. UV absorbance, detection limits of 10-80

A solid coated device is disclosed that has covalently attached a coating which comprises a first layer contg. first functional groups, to which first layer functional groups is covalently attached a second, linking layer carrying second functional groups, to which second, linking layer is covalently attached a third, proximal phospholipid layer, to which third, proximal phospholipid layer is noncovalently attached a fourth, distal lipid layer, so that the proximal and the distal lipid layers together form

Time-resolved fluorescence anisotropy of the single tryptophan (Trp) residue of melittin was measured to assess the orientational fluctuations of the protein in lipid membranes above and below the lipid phase transition temp. (Tt). In phospholipid vesicles contg. melittin, 3 relaxation times were obsd. at temps. above Tt, whereas only 2 relaxation times were obsd. below Tt; the slowest of the 3 relaxations in fluid membranes was absent in the ordered membranes. In planar multibilayer membranes at temps. abo

A review with 11 refs. on a fluorescent Aib-contg. peptide model for the acetylcholine receptor ion channel. This peptide induces single-channel conductance in planar lipid bilayers exhibiting one single open conductance state and weakly voltage-dependent kinetics. The peptides fluorescent dansyl group can be used to correlate motions of the peptide helix with channel opening and closing. [on SciFinder (R)]
