Mitochondrial stress requires timely intervention to prevent mitochondria' and cellular dysfunction. Re-establishing the correct protein homeostasis is crucial for coping with mitochondria' stress and maintaining cellular homeostasis. The best-characterize ...

Sirt1 is an NAD1-dependent deacetylase that extends lifespan in lower organisms and improves metabolism and delays the onset of age-related diseases in mammals. Here we show that SRT1720, a synthetic compound that was identified for its ability to activate ...

We highlight papers by Diaz-Cuadros et al.(1) and Iwata et al.(2) that demonstrate the role of mitochondrial metabolism in setting developmental pace through their control over cellular bioenergetics and redox homeostasis in mice and humans. ...

Mutations in nuclear-encoded mitochondrial genes are responsible for a broad spectrum of disorders among which Leigh syndrome is the most common in infancy. No effective therapies are available for this severe disease mainly because of the limited capabili ...

AMP-activated protein kinase (AMPK) is a metabolic fuel gauge conserved along the evolutionary scale in eukaryotes that senses changes in the intracellular AMP/ATP ratio. Recent evidence indicated an important role for AMPK in the therapeutic benefits of m ...

Fast, label-free, high-resolution, three-dimensional imaging platforms are crucial for high-throughput in vivo time-lapse studies of the anatomy of Caenorhabditis elegans, one of the most commonly used model organisms in biomedical research. Despite the ne ...

Little information is available about how post-transcriptional mechanisms regulate the aging process. Here, we show that the RNA-binding protein Pumilio2 (PUM2), which is a translation repressor, is induced upon aging and acts as a negative regulator of li ...

Highlights • LPCAT3 and ELOVL5 coordinate PUFA metabolism in macrophages • Dual LPCAT3/ELOVL5 deletion increases membrane cholesterol-enriched domains • This alteration enhances macrophage sensitivity to cytotoxic oxysterols ...

NAD(+) has emerged as a vital cofactor that can rewire metabolism, activate sirtuins, and maintain mitochondrial fitness through mechanisms such as the mitochondrial unfolded protein response. This improved understanding of NAD(+) metabolism revived intere ...

The present invention relates to agents that prevent or reverse process of stem cell senescence. Further, the invention relates to methods and compositions useful in the prevention and/or treatment of stem senescence. ...

Brown adipose tissue (BAT) has abundant mitochondria with the unique capability of generating heat via uncoupled respiration. Here, Park et al. identify LETMD1 as a mitochondrial matrix protein enriched in brown adipose tissue (BAT) and reveal a crucial ro ...

Mitochondria have emerged as key actors of innate and adaptive immunity. Mitophagy has a pivotal role in cell homeostasis, but its contribution to macrophage functions and host defense remains to be delineated. Here, we showed that lipopolysaccharide (LPS) ...

Bile acids are increasingly being appreciated as complex metabolic integrators and signalling factors and not just as lipid solubilizers and simple regulators of bile-acid homeostasis. It is therefore not surprising that a number of bile-acid-activated sig ...

The present invention provides methods of treating various disorders associated with mitochondrial dysfunction, including but not limited to metabolic disorders, neurodegenerative diseases, chronic inflammatory diseases, and diseases of aging. ...

The invention relates generally to methods, agents and pharmaceutical compositions for treating or preventing an aging-related disease by reducing the levels and/or activity of the RNA-binding protein PUM2. ...

The present invention relates to novel biomarkers for aging and healthspan, methods and uses thereof, in particular, for determining the biological age of a subject and/or preventing or delaying aging process in a subject, and kits for use in said methods. ...

Spatiotemporally regulated targeted gene manipulation is a common way to study the effect of gene variants on phenotypic traits, but the Cre/loxP and Tet-On/Tet-Off systems can affect whole-organism physiology and function due to off-target effects. We hig ...
