Stem cells reside in complex niches in which their behaviour is tightly regulated by various biochemical and biophysical signals. In order to unveil some of the crucial stem cell-niche interactions and expedite the implementation of stem cells in clinical ...

The synthesis of novel hybrid hydrogels by stepwise copolymn. of multi-arm vinyl sulfone-terminated poly(ethylene glycol) macromers and a-w cysteine oligopeptides via Michael-type addns. is described. Crosslinking kinetics, studied by in situ rheometry, can be controlled by pH and the presence of charged amino acid residues in close proximity to the Cys, which modulates the pKa of the thiol group. These end-linked networks were characterized by their equil. swelling in water, by their viscoelastic propertie

Gastruloids are 3D structures generated from pluripotent stem cells recapitulating fundamental principles of embryonic pattern formation. Using single-cell genomic analysis, we provide a resource mapping cell states and types during gastruloid development ...

Lab-grown 'mini-colons' enable the simulation of colorectal tumor dynamics and the high-resolution study of interactions with cells from the tumor's native microenvironment. These next-generation organoids offer a wealth of new experimental opportunities, ...

By binding growth factors (GFs), the ECM tightly regulates their activity. We recently reported that the heparin-binding domain II of fibronectin acts as a promiscuous high-affinity GF-binding domain. Here we hypothesized that fibrin, the provisional ECM d ...

Computer-controlled hydrodynamic flow focusing was utilized to generate tethered protein gradients of any user-defined shape on the surface of soft synthetic hydrogels. ...

Extra-cellular matrix (ECM) tethered morphogen gradients play an important role in tissue development and regeneration. There is currently only a limited set of model systems which can mimic these gradients in an artificial ECM-like environment. We have de ...

Microwell arrays are cell culture and imaging platforms to assess cells at a single cell level and in high-throughput. They allow the spatial confinement of single cells in microfabricated cavities on a substrate and thus the continuous long-term observation of single cells and their progeny. The recent development of microwell arrays from soft, biomimetic hydrogels further increases the physiological relevance of these platforms, as it substantially enhances stem cell survival and the efficiency of self-re

T cell development depends critically on the thymic stroma—in particular, the diverse array of functionally distinct thymic epithelial cell (TEC) types. However, a robust in vitro thymus model mimicking the native thymus and compatible with medium-/high-th ...

Fate choices of stem cells are regulated in response to a complex array of biochemical and physical signals from their microenvironmental niche. Whereas the molecular composition and the role of mechanical niche cues have been extensively studied, relative ...

Building complex tissues requires the development of innovative interdisciplinary engineering solutions. In this Forum, the INTENS Consortium discuss experimental considerations and challenges for generating a tissue-engineered intestine for the treatment ...

Haematopoietic stem cells (HSCs) differ from their committed progeny by relying primarily on anaerobic glycolysis rather than mitochondrial oxidative phosphorylation for energy production. However, whether this change in the metabolic program is the cause ...

The recent increase in organoid research has been met with great enthusiasm, as well as expectation, from the scientific community and the public alike. There is no doubt that this technology opens up a world of possibilities for scientific discovery in de ...

Fatty acid beta-oxidation (FAO), the breakdown of lipids, is a metabolic pathway used by various stem cells. FAO levels are generally high during quiescence and downregulated with proliferation. The endogenous metabolite malonyl-CoA modulates lipid metabol ...

Glandular epithelia, including mammary gland (MG) and prostate, are composed of luminal and basal cells. During embryonic development, glandular epithelia arise from multipotent stem cells (SCs) that are replaced after birth by unipotent basal and unipoten ...

Research in developmental biology has been recently enriched by a multitude of in vitro models recapitulating key milestones of mammalian embryogenesis. These models obviate the challenge posed by the inaccessibility of implanted embryos, multiply experime ...

The treatment of bone defects with recombinant bone morphogenetic protein-2 (BMP-2) requires high doses precluding broad clinical application. Here, a bioengineering approach is presented that strongly improves low-dose BMP-2-based bone regeneration by mob ...

As the field of tissue engineering develops, methods for screening combinations of signals for their effects on stem cell behavior are needed. We introduce a microgel-based screening platform for testing combinations of in situ-generated proteins on stem c ...

By using light to control the degradation of hydrogel components in space and time, researchers have generated a tool to help them reconstruct functional biological tissues in a culture dish. ...

Liver organoids have emerged as promising in vitro models for toxicology, drug discovery, and disease modeling. However, conventional 3D epithelial organoid culture systems suffer from significant drawbacks, including limited culture duration, a nonphysiol ...
