Epithelial organoids are stem cell-derived tissues that approximate aspects of real organs, and thus they have potential as powerful tools in basic and translational research. By definition, they self-organize, but the structures formed are often heterogen ...

Existing organoid models fall short of fully capturing the complexity of cancer because they lack sufficient multicellular diversity, tissue-level organization, biological durability and experimental flexibility. Thus, many multifactorial cancer processes, ...

Proper tissue function and regeneration rely on robust spatial and temporal control of biophysical and biochemical microenvironmental cues through mechanisms that remain poorly understood. Biomaterials are rapidly being developed to display and deliver ste ...

Mechanisms controlling the proliferative activity of neural stem and progenitor cells (NSPCs) have a pivotal role to ensure life-long neurogenesis in the mammalian brain(1). How metabolic programs are coupled with NSPC activity remains unknown. Here we sho ...

Biomolecular signaling is of utmost importance in governing many biological processes such as the patterning of the developing embryo where biomolecules regulate key cell-fate decisions. In vivo, these factors are presented in a spatiotemporally tightly co ...

Clinical translation of engineered tissues into regenerative medicine applications, and the effort to reduce the use of animals for the screening of drugs and other compounds, result in an increasing demand for human tissues engineered in vitro for implant ...

Cells operate as networks of proteins, membranes, condensates and compartments, each sensing and displaying distinct intracellular mechanical properties. However, cells also sense, adapt and respond to manifold mechanical properties of the environment, including adhesion, tension, stiffness, shear, viscoelasticity, plasticity, pressure and confinement. By gauging these properties at various timescales and across nano to macro length scales, cellular systems alter their collective responses. The field of mec

Advances in organoid models, as ex vivo mini-organs, and the development of screening imaging technologies have continuously driven each other forward. A complete understanding of organoids requires detailed insights into the intertwined intraorganoid and ...

Despite increasingly stringent methods to isolate hematopoietic stem cells (HSCs), considerable heterogeneity remains in terms of their long-term self-renewal and differentiation potential. Recently, the existence of long-lived, self-renewing, myeloid-rest ...

Engineered artificial microenvironments hold enormous potential as models to study developmental, physiological, pathological, and regenerative processes under highly defined conditions. Such platforms aim at bridging the gap between traditional in vitro 2 ...

Freshly isolated muscle stem cells (MuSCs) exhibit robust regenerative capacity in vivo that is rapidly lost in culture. Using a bioengineered substrate to recapitulate key biophysical and biochemical niche features in conjunction with a novel highly autom ...

The emergence of multiple axes is an essential element in the establishment of the mammalian body plan. This process takes place shortly after implantation of the embryo within the uterus and relies on the activity of gene regulatory networks that coordina ...

Reductionist in vitro model systems which mimic specific extracellular matrix functions in a highly controlled manner, termed artificial extracellular matrices (aECM), have increasingly been used to elucidate the role of cell-ECM interactions in regulating ...

Inkjet printing is widely considered a promising strategy to pattern hydrogels and living cells into three-dimensional (3D) constructs that structurally resemble tissues in our body. However, this approach is currently constrained by the limited control ov ...

Engineering synthetic hydrogels on a molecular basis to introduce natural features that are important in instructing cell behavior is becoming increasingly crucial in biomaterial-based approaches for regenerative medicine and in cell biology to study cell– ...

Changes in stem cell activity may underpin aging. However, these changes are not completely understood. Here, we combined single-cell profiling with machine learning and in vivo functional studies to explore how hematopoietic stem cell (HSC) divisions patt ...

Organoids have stepped into the limelight as unique in vitro systems for modeling organ development, function and disease. This review provides a perspective on how chemically defined, bio-inspired hydrogels could be used for replacing ill-defined matrices ...

The potential of stem cells in clinics and as a diagnostic tool is still largely unmet, partially due to a lack of in vitro models that efficiently mimic the in vivo stem cell microenvironment—or niche—and thus would allow reproducible propagation of stem cells or their controlled differentiation in vitro. The current methodological challenges in studying and manipulating stem cells have spurred intense development and application of microfabrication and micropatterning technologies in stem cell biology. Th

Due to their selective crosslinking chemistry and well-defined mechano-biochemical characteristics, hydrogels crosslinked by thiol-Michael addition reactions have garnered immense interest for 3D cell culture applications. These hydrogels are typically for ...

Stem cells are characterized by their dual ability to self- renew and differentiate, yielding essentially unlimited numbers of progeny that can replenish tissues with either high turnover such as blood and skin or contribute to the regeneration of organs with less frequent remodeling such as muscle. In contrast to their embryonic counterparts, adult stem cells can only preserve their unique functions if they are in intimate contact with an instructive microenvironment, termed niche. Stem cells integrate a c
