Supported cell-membrane sheets are promising in vitro systems to investigate the properties of membranes with native protein/lipid composition, in particular their sub-compartmentalization and the differential localization of proteins associated to them. W ...

Structure and orientation of mols. are key properties of functionalized surfaces. Using time-of-flight secondary ion mass spectrometry (TOF-SIMS), here we investigate how to modulate these parameters upon the immobilization process varying the conditions of self-assembly. The mol. of interest, a template-assembled synthetic protein (TASP), consists of a central peptide ring with orthogonally arranged residues. Thioalkane chains allow the directed self-assembly of the mol. on a gold surface; four serine resi

Method and apparatus to study biological molecules or biological components at or in self-assembled monolayers (SAMs) on metal surfaces (2) such as gold in aqueous environments (6) using attenuated total internal reflection Fourier transform infra-red (ATR-FTIR) spectroscopy are described. Said method may be used for e.g. the screening for effective substances suitable as agonists or antagonists or - if performed time resolved - for studying kinetics of interactions of e.g. biological molecules adsorption a

A dry sorption system for F removal from kiln flue gases using CaCO3 (limestone) produces flue gases with a residual F content of =21 days in the reactor. The flue gases are taken from the kiln firing process and passed to the reaction cell at a flow velocity of

Time-resolved fluorescence anisotropy (FA) measurements are reported for 5 helical, bilayer-spanning heneicosapeptides, each contg. 1 tryptophan (Trp) at sequence positions 1, 6, 11, 16, and 21, resp. The FA decay reflected 2 mol. processes in all cases, local internal fluctuations of the Trp side-chain with a relaxation time of 200-500 ps and motions of the whole polypeptide mol. with a relaxation time of 9-10 ns. Concomitant mol. dynamics calcns. indicated a correlation between the fast FA decay and confo

The in vivo and in vitro labeling of fusion proteins with synthetic mols. capable of probing and controlling protein function has the potential to become an important method in functional genomics and proteomics. We have recently introduced an approach for the specific labeling of fusion proteins, which is based on the generation of fusion proteins with the human DNA repair protein O6-alkylguanine-DNA alkyltransferase (hAGT) and the irreversible reaction of hAGT with O6-benzylguanine derivs. Here, we report

We discuss the successful transport of jurkat cells and 5D10 hybridoma cells using a reciprocating micropump with nozzle-diffuser elements. The effect of the pumping action on cell viability and proliferation, as well as on the damaging of cellular membran ...

A review with 130 refs. on optical techniques such as total internal reflection fluorescence, waveguides, and SPR for the detection of mol. interactions of membrane receptors. The outlook on how these emerging techniques may be applied to solve important questions in the field of mol. membrane biol. is given. [on SciFinder (R)]

CD8+ cytotoxic T lymphocyte (CTL) can recognize and kill target cells that express only a few cognate major histocompatibility complex class I-peptide (pMHC) complexes. To better understand the mol. basis of this sensitive recognition process, the authors ...

A bifacial topological template exhibiting metal-binding sites and thioalkane chains has been incorporated into self-assembled monolayers and immobilised on gold surfaces: These systems allow for the detection of external ligands by SPR spectroscopy representing a first step in developing biosensors based on the TASP (Template Assembled Synthetic Proteins) concept.

A review with 23 refs. about detn. of conformation of membrane proteins, e.g., melittin, by Raman-spectroscopy. [on SciFinder (R)]

After decades of slow progress, the pace of research on membrane protein structures is beginning to quicken thanks to various improvements in technology, including protein engineering and microfocus X-ray diffraction. Here we review these developments and, ...

CdSe quantum dots (QDs) with a high fluorescence quantum yield of 25% and a narrow size distribution were synthesized in a single step in H2O using glutathione as a stabilizing mol. The exceptional optical properties enabled for the 1st time the detection of in-H2O-prepd. single quantum dots at room temp. For application as fluorescent bioanal. probes, the QDs were coated with streptavidin. These QDs self-assemble with high contrast on micropatterned biotin while preserving their optical properties and thei

A chimeric TASP (Template Assembled Synthetic Proteins) mol., consisting of a 4-helix bundle motif and an antibody recognition site, has been covalently attached to a gold surface. The reversible antibody binding to the self-assembled monolayer as monitored by surface plasmon resonance spectroscopy demonstrates the versatility of the TASP concept for the construction of biosensors. [on SciFinder (R)]

Nanometer light source: Interaction between high-affinity binding sites on lipid-coated nanocrystals (NCs) and different proteins enables the controlled formation of supramol. nanostructures on micropatterned surfaces. The NCs serve as local light sources ...

With the possibility of creating and handling nanometer-sized objects, it became popular to dream of future miniaturized tools allowing completely new applications in medicine and technology. Mother nature created during evolution many such self-assembled highly sophisticated and robust objects, and it is very tempting to modify them for other purposes: artificial cells as intelligent submarines floating in the bloodstream attacking cancer cells, single-molecule detection and manipulation allowing the botto

The authors probed the function of receptor protein OmpF by in situ measurement of both the binding of ligands by surface plasmon resonance (SPR) and subsequent changes of the channel activity by impedance spectroscopy (IS). [on SciFinder (R)]

We report on the fabrication and electrical characterization of functionalized solid-state nanopores in low stress silicon nitride membranes. First, a pore of approximately 50 nm diameter was drilled using a focused ion beam technique, followed by the loca ...

The hybridoma cell line ZAC3 expresses Vibrio cholerae lipopolysaccharide (LPS)-specific mouse IgA mols., as a heterogeneous population of monomeric (IgAm), dimeric (IgAd), and polymeric (IgAp) forms. We describe a gentle method combining ultrafiltration, ion-exchange chromatog., and size exclusion chromatog. for the simultaneous and qual. sepn. of the three mol. forms. Milligram quantities of purified IgA mols. were recovered allowing for direct comparison of the biol. properties of the three forms. LPS bi

A method is presented to form gold-electrode-tethered lipid bilayers with exceptionally high elec. resistances. Elec. impedance spectroscopy is used to monitor the bilayer incorporation of a ligand-gated ion channel protein and the modulation of its channel activity by the selective binding of an antibody. Due to the low defect d. of the tethered membrane, the effect of a few channels can be resolved thus opening the way to single-channel expts. on this highly stable and versatile platform. In turn, a minut
