Mifepristone alone and in combination with scAAV9-SMN1 gene therapy improves disease phenotypes in Smn2B/- spinal muscular atrophy mice

Spinal muscular atrophy (SMA) is a neuromuscular disease caused by deletions or mutations in the survival motor neuron 1 ( SMN1 ) gene. SMA is characterised by alpha motor neuron loss in the spinal cord and subsequent muscle atrophy. There are currently three approved SMN-directed therapies for SMA patients. While these therapies have transformed what was once a life-limiting condition into one that can be managed and even improved, they are unfortunately not cures, highlighting the need for additional supp

Stress-dependent responses of grapevine wood and fungal pathogen activity under esca and drought

Summary Biotic and abiotic stresses impact perennial plants, altering plant physiology with consequences on fungal endophytes and disease expression. In grapevine, one of the world’s most valuable crops, drought inhibits esca disease expression, but the underlying plant and fungi molecular interactions are unknown. We integrated wood metatranscriptomics, metabolomics, and metabarcoding to investigate such interactions in 30 year-old grapevine and eight key wood pathogenic fungi under drought or esca leaf sy

Wastewater and green waste biorefinery Degradation of simple sugars using mixed cultures of purple non-sulfur bacteria: an insight into microbial interactions

Purple non-sulfur bacteria (PNSB) are phototrophic organisms that have raised interest for both industrial and domestic wastewater treatment and valorization because of their very high assimilation (~1 g CODX g-1 COD substrate) and valuable compounds production (i.e hydrogen, polyhydroxyalkanoates, pigments). PNSB have been studied mostly in pure- or co-cultures with fermentative bacteria. Mixed cultures have been little described. This work aims to enrich a stable association of fermenters and PNSB on a ca

Enzyme annotation for orphan and novel reactions using knowledge of substrate reactive sites

Vassily Hatzimanikatis, Ljubisa Miskovic, Noushin Hadadi, Marianne Seijo

Thousands of biochemical reactions with characterized activities are “orphan,” meaning they cannot be assigned to a specific enzyme, leaving gaps in metabolic pathways. Novel reactions predicted by pathway-generation tools also lack associated sequences, l ...

Benzimidazole derivative small-molecule 991 enhances AMPK activity and glucose uptake induced by AICAR or contraction in skeletal muscle

Caterina Collodet

AMP-activated protein kinase (AMPK) plays diverse roles and coordinates complex metabolic pathways for maintenance of energy homeostasis. This could be explained by the fact that AMPK exists as multiple heterotrimer complexes comprising a catalytic alpha-s ...

Functional characterization of a new tetrachloroethene reductive dehalogenase of Sulfurospirillum spp. identified in mixed cultures

Christof Holliger

Recent analysis of a set of reductive dehalogenases revealed that sequence similarity and substrate specificity are not correlated, making functional prediction from sequences difficult (1). To this respect, several enrichment cultures were investigated for their distinct dechlorination potential of tetrachloroethene (PCE). Indeed, SL2-PCEb, an enrichment culture dominated by Sulfurospirillum spp., was shown to stepwise dechlorinate PCE to TCE and cis-DCE, suggesting the successive involvement of two dechlo

Sirtuins as regulators of metabolism and healthspan

Since the beginning of the century, the mammalian sirtuin protein family (comprising SIRT1-SIRT7) has received much attention for its regulatory role, mainly in metabolism and ageing. Sirtuins act in different cellular compartments: they deacetylate histones and several transcriptional regulators in the nucleus, but also specific proteins in other cellular compartments, such as in the cytoplasm and in mitochondria. As a consequence, sirtuins regulate fat and glucose metabolism in response to physiological c

Theoretical treatment and numerical simulation of potentiometric and amperometric enzyme electrodes and of enzyme reactors. Part 2: Time-dependent concentration profiles, fluxes, and responses

Nico de Rooij

The time-dependent behavior of sensor systems based on enzyme membranes is treated theoretically. The comparative treatment covers traditional potentiometric and amperometric enzyme electrodes as well as enzyme reactors. Theoretical solutions are derived by the method of Laplace transforms. The relationships refer to the analytically useful range of relatively low substrate concentrations where the response signal is a function of the sample concentration. Numerical simulations using finite-difference proce

Glial Glycogen Metabolism

Glycogen represents the largest energy reserve of the brain and is almost quite exclusively localized in astrocytes. In the first part of this article, a summary of our knowledge concerning brain glycogen metabolism and its regulation is presented. The second part focuses on potential evidence that glycogen can serve as energy substrate during neuronal activation, as well as on the role of brain glycogen as an emergency reserve in case of insufficient energy supply. These data are discussed in reference to
