Gating of an artificial receptor was monitored for the first time by impedance spectroscopy in tethered lipid bilayer. Subnanomolar concns. of antibody were detected by modulation of the channel activity. The principle of gating by an antibody is particularly attractive because of its general character, and thus opens new possibility of immunosensing. [on SciFinder (R)]

Many central biological events rely on protein-ligand interactions. The identification and characterization of protein-binding sites for ligands are crucial for the understanding of functions of both endogenous ligands and synthetic drug molecules. G prote ...

Structural biology has come a long way, from being a niche interest of specialists to where we are today, with over 127,500 biomolecular structures available in the Protein Data Bank (PDB) as of the writing of this letter. The two main factors that have be ...

Dansyl-(Ala-Aib-Ala-Aib-Ala)3-Pro-Ala-Aib-Ala-Aib-Ala-Trp-OMe [(I), where Aib is a-aminoisobutryate] is a bent a-helical peptide which displays ion channel activity characteristic of the nicotinic acetylcholine receptor. The membrane organization, side-chain and backbone motions, and voltage dependence of the I ion channel are discussed. [on SciFinder (R)]

Fluorescence resonance energy transfer (FRET) is a powerful technique to reveal interactions between membrane proteins in live cells. Fluorescence labeling for FRET is typically performed by fusion with fluorescent proteins (FP) with the drawbacks of a lim ...

Alkanethiolates have been widely used as chemisorbates to modify gold surfaces, in spite of their relatively poor oxidative stability. We introduce gold-chemisorbing block copolymers bearing an anchoring block of poly(propylene sulfide) (PPS), selected in the expectation of greater stability. These materials offer a more robust approach to surface modification of gold. As an example, a triblock copolymer with poly(ethylene glycol) (PEG) was selected, with the goal of minimizing biol. adsorption and adhesion

High-d. microarrays of attoliter vol. elements can be created within minutes in a parallel and effortless manner by using self-assembly of nanometer-size components based on biol. recognition. These ultrasmall vols. allow localization of single mols., screening for (bio)chem. properties, and assessing the performance of confined chem. reactions. [on SciFinder (R)]

A review and discussion with 5 refs., on membrane insertion of bacteriophage M13 procoat protein, giving emphasis to structural fluctuations during assocn. with membranes. [on SciFinder (R)]

The authors have investigated the interaction of fluorescent peptide ligands with the G protein-coupled receptor NK2 using novel spectrofluorometric approaches. Several heptapeptide antagonists of structure PhCO-Xaa-Ala-D-Trp-Phe-D-Pro-Pro-Nle-NH2 were labeled on position 1 (Xaa) with the environment-sensitive nitrobenzoxadiazole (NBD) probe, differing only in the length of the spacer between the NBD group and the peptide. Upon binding of the labeled antagonist to NK2 receptors stably expressed in Chinese h

The authors report here thermodn. of the membrane insertion process of the M13 procoat protein, a lipid bilayer traversing protein comprising a leader sequence. [on SciFinder (R)]

A micrometer-sized affinity bead (red) is (i) taken up into a cell by phagocytosis, (ii) photochemically released from phagosomes, (iii) optically trapped by the cell, and (iv) isolated by cell lysis for subsequent analysis of captured intracellular analyt ...
