The binding of metal ions to self-assembled nitrilotriacetic acid (NTA)-modified thioalkane monolayers was monitored by impedance spectroscopy via a capacitance change of the blocking gold electrode. Binding to a fluorescent NTA-derivative and the native NTA, measured in bulk by fluorescence quenching and isothermal titration calorimetry, was determined for comparison. Surface and bulk absolute dissociation constants K-abs of metal ion-NTA complexes for these three derivatives showed no significant differen

We have expressed the 5-HT3 receptor with two transient expression sytems: the Semliki Forest virus and the HEK293EBNA. Both systems allow the rapid expression of functional receptor protein at very high level: up to 107 receptors per cell. Large scale production yielded 1 to 2 mg of receptor protein per liter. The solubilisation of the receptor from membranes has been optimised; C12E9 solubilised 50 to 60 % of the ligand binding activity. A C-terminally engineered hexahistidine-tag facilited efficient puri

CdSe quantum dots (QDs) with a high fluorescence quantum yield of 25% and a narrow size distribution were synthesized in a single step in H2O using glutathione as a stabilizing mol. The exceptional optical properties enabled for the 1st time the detection of in-H2O-prepd. single quantum dots at room temp. For application as fluorescent bioanal. probes, the QDs were coated with streptavidin. These QDs self-assemble with high contrast on micropatterned biotin while preserving their optical properties and thei

Nanometer light source: Interaction between high-affinity binding sites on lipid-coated nanocrystals (NCs) and different proteins enables the controlled formation of supramol. nanostructures on micropatterned surfaces. The NCs serve as local light sources ...

We are introducing a new approach to evaluate cellular uptake of drugs and drug candidates into living cells. The approach is based on converting the protein target of a given class of compounds into a fluorescent biosensor. By measuring the binding of dif ...

We present a method probing the local membrane environment surrounding membrane proteins in the plasma membrane by linking a solvatochromic dye to the target, and provide evidence for lipid-based sorting. The insulin receptor environment changed upon insul ...

Cellular signaling is classically investigated by measuring optical or electrical properties of single or populations of living cells. Here we show that ligand binding to cell surface receptors and subsequent activation of signaling cascades can be monitor ...

A fluorogenic probe for plasma membrane proteins based on the dye Nile Red and SNAP-tag is introduced. It takes advantage of Nile Red, a solvatochromic molecule highly fluorescent in an apolar environment, such as cellular membranes, but almost dark in a p ...

Fluorescence resonance energy transfer (FRET) is a powerful technique to reveal interactions between membrane proteins in live cells. Fluorescence labeling for FRET is typically performed by fusion with fluorescent proteins (FP) with the drawbacks of a lim ...

Repression of endogenous retroviral elements (ERVs) is facilitated by KAP1 (KRAB-associated protein 1)-containing complexes, however the underlying mechanism remains unclear. Here, we show that binding of KAP1 to the major component of the heterochromatin ...

A micrometer-sized affinity bead (red) is (i) taken up into a cell by phagocytosis, (ii) photochemically released from phagosomes, (iii) optically trapped by the cell, and (iv) isolated by cell lysis for subsequent analysis of captured intracellular analyt ...
