Résumé

The production of reactive oxygen species (ROS) in normal tissues is associated with side effects of many cancer treatments. This is, in particular, the case for the skin photosensitization experienced by certain patients subjected to PDT based on a systemic administration of photosensitizers (PS). Similarly, the ROS generated in normal tissues during radiotherapies are responsible for the damages induced in various organs, including the skin.

Thérapie photodynamique

La thérapie photodynamique (ou photothérapie dynamique ; photochimiothérapie ; en photodynamic therapy, abrégée en PDT), est une technique de traitement médical local et sélectif visant à détruire des tissus pathologiques, ou certains microbes, grâce à l'utilisation de substances chimiques photosensibilisantes devenant cytotoxiques une fois activés par de une lumière (pour une gamme réduite de longueurs d'onde) ou dans certains cas par des rayons X (qui peuvent pénétrer en profondeur dans l'organisme).

Photosensitizer

Photosensitizers are light absorbers that alters the course of a photochemical reaction. They usually are catalysts. They can function by many mechanisms, sometimes they donate an electron to the substrate, sometimes they abstract a hydrogen atom from the substrate. At the end of this process, the photosensitizer returns to its ground state, where it remains chemically intact, poised to absorb more light. One branch of chemistry which frequently utilizes photosensitizers is polymer chemistry, using photosensitizers in reactions such as photopolymerization, photocrosslinking, and photodegradation.

Treatment of cancer

Cancer can be treated by surgery, chemotherapy, radiation therapy, hormonal therapy, targeted therapy (including immunotherapy such as monoclonal antibody therapy) and synthetic lethality, most commonly as a series of separate treatments (e.g. chemotherapy before surgery). The choice of therapy depends upon the location and grade of the tumor and the stage of the disease, as well as the general state of the patient (performance status). Cancer genome sequencing helps in determining which cancer the patient exactly has for determining the best therapy for the cancer.
