When many (m) null hypotheses are tested with a single dataset, the control of the number of false rejections is often the principal consideration. Two popular controlling rates are the probability of making at least one false discovery (Bonferroni) and the expected fraction of false discoveries among all rejections (Benjamini-Hochberg). Methods controlling these rates based on the ordered p-values p(1) , ... p(m) are well-known. In this talk, we present a new family of multiple testing procedures that brid

Background: The growth of tumor cells is accompanied by mutations in nuclear and mitochondrial genomes creating marked genetic heterogeneity. Tumors also contain non-tumor cells of various origins. An observed somatic mitochondrial mutation would have occu ...

A method is described to discover if a gene carries one or more allelic mutations that confer risk for any specified common disease. The method does not depend upon genetic linkage of risk-conferring mutations to high frequency genetic markers such as single nucleotide polymorphisms. Instead, the sums of allelic mutation frequencies in case and control cohorts are determined and a statistical test is applied to discover if the difference in these sums is greater than would be expected by chance. A statistic

Background Colorectal cancer (CRC) is the second leading cause of cancer-related deaths worldwide. CRC deaths can be reduced with prevention and early diagnosis. Circulating tumor DNA-based liquid biopsies, are emerging tools for cancer detection. However, ...

John Wilder Tukey, Donner Professor of Science Emeritus at Princeton University, was born in New Bedford, Massachusetts, on June 16, 1915. After earning bachelor's and master's degrees in chemistry at Brown University in 1936 and 1937, respectively, he started his career at Princeton University with a Ph.D. in mathematics in 1939 followed by an immediate appointment as Henry B. Fine Instructor in Mathematics. A decade later, at age 35, he was advanced to a full professorship. He directed the Statistical Res

We study bias arising as a result of nonlinear transformations of random variables in random or mixed effects models and its effect on inference in group-level studies or in meta-analysis. The findings are illustrated on the example of overdispersed binomi ...

Gaia is a cornerstone mission in the science programme of the European Space Agency (ESA). The spacecraft construction was approved in 2006, following a study in which the original interferometric concept was changed to a direct-imaging approach. Both the ...

Detecting local differences between groups of connectomes is a great challenge in neuroimaging, because the large number of tests that have to be performed and the impact on multiplicity correction. Any available information should be exploited to increase ...

We have studied the retention indices of 127 volatile substances on a C-78-paraffin and on its seven nearly isochor and isomorphous polar derivatives over a temperature range of 90-210 A degrees C. The retention index of a substance on the C-78 paraffin ha ...

Context. At about 1000 days after the launch of Gaia we present the first Gaia data release, Gaia DR1, consisting of astrometry and photometry for over 1 billion sources brighter than magnitude 20.7. Aims. A summary of Gaia DR1 is presented along with illu ...

Resting-state functional MRI (rs-fMRI) opens a window on large-scale organization of brain function. However, establishing relationships between resting-state brain activity and cognitive or clinical scores is still a difficult task, in particular in terms ...

In multiple testing, a variety of control metrics of false positives have been introduced such as the Per Family Error Rate (PFER), Family-Wise Error Rate (FWER), the False Discovery Rate (FDR), the False Exceedence Rate (FER). In this talk, we present a comprehensive family of error rates together with a corresponding family of multiple testing procedures (MTP). Based on the needs of the problem at hand, the user can choose a particular member among these MTPs. The new error rate limits the number of false

Carcinogenesis is commonly described as a multistage process, in which stem cells are transformed into cancer cells via a series of mutations. In this article, we consider extensions of the multistage carcinogenesis model by mixture modeling. This approach ...

Variance stabilization is a simple device for normalizing a statistic. Even though its large sample properties are similar to those of studentizing, many simulation studies of confidence interval procedures show that variance stabilization works better for ...

The ability to identify reliably a positive or negative partial correlation between the expression levels of two genes is determined by the number p of genes, the number n of analyzed samples, and the statistical properties of the measurements. Classical statistical theory teaches us that the product of the root sample size multiplied by the size of the partial correlation is the crucial quantity. But this has to be combined with some adjustment for multiplicity depending on p, which makes the classical ana

We study an adaptive statistical approach to analyze brain networks represented by brain connection matrices of interregional connectivity (connectomes). Our approach is at a middle level between a global analysis and single connections analysis by conside ...

Context. Parallaxes for 331 classical Cepheids, 31 Type II Cepheids, and 364 RR Lyrae stars in common between Gaia and the HIPPARCOS and Tycho-2 catalogues are published in Gaia Data Release 1 (DR1) as part of the Tycho-Gaia Astrometric Solution (TGAS). Ai ...

When testing many null hypotheses, deciding which of them to reject is a subtle game. The prevailing approach consists in deciding on a level and type of control against false rejections (errors of type I) and subject to this constraint to maximize the number of rejections. The two extreme types of control are the FWER (family-wise error rate, probability of at least one false rejection) and FDR (false discovery rate, expected rate of false rejections among all rejections). In this talk, I will discuss two

Over the last two decades, a large variety of type I error rates and control procedures have been proposed in the field of multiple hypotheses testing. This paper proposes a framework that includes many existing proposals by investigating procedures in whi ...
