Yeast strains were examd. for their ability to produce glycerol, and Kluyveromyces marxianus NRRLY-665 was chosen for further optimization due to its comparatively high growth rate (0.15/h) and glycerol yield (9.5% by wt. on lactose) at 37 Deg and pH 7. Optimal conditions for glycerol formation were 30-37 Deg and pH 6-7. Increasing whey permeate concn. above the equiv. of 4.5% lactose decreased growth rate and cell prodn. and increased HOAc prodn. When SO32- was added throughout the fermn. to complex AcH be

The press-cake (PC) of oleaginous species (Brassica napus, Moringa oleifera, Glycine max) has been investigated as a biosorbent for metals removal from contaminated water. ...

In order to extend the shelf life of refrigerated dough, a new method based on thermo-gelling immobilization is proposed. This slows fermn. at low temps. (

The high-pressure freezing (HPF) technique was applied to the cryo-immobilization of alginate gels and the quality of the freezing analyzed on a TEM by comparison of the segregation pattern of samples of decreasing thickness. Dynamic simulations of heat transfer within an idealized slab of pure water surrounded by two walls of aluminum were performed to illustrate the effect of the heat-transfer coeff. by convection on the cooling rate of the sample. Heat-transfer coeffs. in liq. nitrogen and liq. propane a

The (B) over bar (0)(s) -> chi(c2) K+ K- decay mode is observed and its branching fraction relative to the corresponding chi(c1) decay mode, in a +/- 15MeV/c(2) window around the phi mass, is found to be B ((B) over bar (0)(s) -> chi(c2) K+ K-)/B((B) over ...

Animal cell (Chinese Hamster Ovary) concn. was detd. online in a packed bed process using dielec. spectroscopy. This enabled the evaluation of the effect of temp. on specific metabolic rates during 3 mo of continuous culture. The effect of low cultivation temp. on cell growth and metab. was monitored, and the data were used for process development. At 37 DegC cells grew exponentially with a specific growth rate of 0.038 d-1 and specific glucose uptake and lactate prodn. rates increased continually. Redn. of

Accurate online monitoring of biomass is an enduring problem. Of the various methods available, one of the more satisfactory is with dielec. spectroscopy (often referred to as capacitance monitoring). However, as this technique measures total -biovolume', large changes in cellular morphol. or population distribution will alter the calibration with respect to cell wt. or no. In an attempt to minimize this problem, the full dielec. spectrum was scanned every 10-20 min, and then multivariate techniques were us

Monoclonal IgA antibodies have been produced by a hybridoma cell line grown in different bioreactors using serum-contg. and protein-free media in both basal and fortified versions. The effects of culture conditions on IgA prodn. and quality were studied using an anti a-chain ELISA, an antigen-specific ELISA, and anti a-chain Western blotting from which the fractions of the various IgA mol. forms were estd. by densitometry measurements. In stirred-tank reactor (STR) batch cultures, a significant increase in

An amplitude analysis of B-0 ((+-))(K+-) decays is performed in the two-body invariant mass regions 300 < m((+-)) < 1100 MeV/c(2), accounting for the (0), , f(0)(500), f(0)(980) and f(0)(1370) resonances, and 750 < m(K+-) < 1200 MeV/c(2), which is dominate ...

A unique bench-scale heat flux calorimeter is described, which is capable of detecting the low heat signal of an animal cell culture (a max. of 200 mW/l is produced with a batch culture of SF9 insect cells). After the tech. modifications that enhanced the sensitivity of a com. heat flux reaction calorimeter, RC1 from Mettler-Toledo AG. Switzerland, the behavior of this system is analyzed here under biol. operation conditions. Not only the heat flows provoked by the bioprocess parameters largely det. the fin

A single spectra library was used to monitor on-line, by mid-infrared spectroscopy, nine different batch cultures of Escherichia coli performed with various medium compositions, including chemically complex formulations. Whereas the classic chemometrics ap ...

A simple definition of the energetic growth efficiency both in terms of enthalpy and free energy is given and discussed. In cultures growing by respiratory catabolism, these efficiencies may conveniently be obsd. online by isothermal reaction calorimetry. If the fermentative part of catabolism is important, the free energy growth efficiency must be evaluated based on biomass yield measurements. In continuous cultures of 2 yeasts growing by respiration, by pure fermn., and on a mixed catabolism, the efficien

A review with 55 refs. Oxygen is a key substrate in animal cell metab. It has been reported that the oxygen uptake rate (OUR) is a good indicator of cellular activity, and even under some conditions, a good indicator of the no. of viable cells. The measurement of OUR is difficult due to many different reasons. In particular, the very low specific consumption rate (0.2*10-12 mol cell h-1), the sensitivity of the cells to variations in dissolved oxygen concn. and the difficulty to provide oxygen without damag

A search for the rare leptonic decay B +. mu + mu - mu +.mu is performed using proton- proton collision data corresponding to an integrated luminosity of 4.7 fb - 1 collected by the LHCb experiment. The search is carried out in the region where the lowest ...

Heat produced by hybridoma suspension cultures was continuously monitored in a 2-L heat-flux calorimeter. Heat measurements reflected changes in the metabolic state of the culture under different aeration conditions. A highly anaerobic metab. was indicated by the low ratios of heat produced to glucose consumed. [on SciFinder (R)]

The growth kinetics of the sulfate-reducing bacteria Desulfovibrio desulfuricans Essex 6 was investigated under various conditions for potential use in a microbial fuel cell that recovers electrons generated from the redn. of sulfate to hydrogen sulfide. Hydrogen sulfide was found to inhibit growth and decrease both the growth yields and the sulfate-specific redn. rate. Hydrogen sulfide inhibition was direct, reversible, and not due to limitation by iron deficiency. A high initial lactate concn. also retard

A novel bench-scale calorimeter, which is based on a heat-flux principle and modified to accommodate biol. reactions, was evaluated for detg. the heat evolution rate of an aerobic culture of the yeast Kluyveromyces fragilis grown on lactose. The instantaneous heat evolution rate was an accurate indicator for the instantaneous overall metabolic activity. On the other hand, the integrated heat prodn. increased in parallel to the accumulated cell dry wt. The ratios of heat generated per g biomass produced and

Real-time data reconciliation of concentration estimates of process analytes and biomass in microbial fermentations is investigated. A Fourier-transform mid-infrared spectrometer predicting the concentrations of process metabolites is used in parallel with ...

An anti-Rhesus D antibody was produced in various cell-lines and cell-types which were either stable or transient transfected and grown under varying culture conditions. Glycosylation anal. of the IgG1 was carried out mainly by HPAEC methods. The antibody was N-glycosylated only at the conserved glycosylation site at Asn-297 carrying complex type N-glycans as usually present on IgG. The core structure was entirely fucosylated and did not carry intersecting G1cNAc for all antibodies analyzed. Prodn. in SP2/0

Calcium alginate beads were thermally treated at temps. ranging from 25 DegC to 130 DegC for periods of up to 30 min. Important modifications to the structure of the alginate beads were shown to be a function of the temp. and period of incubation at each temp. Modifications to the alginate beads included redn. in size, mech. resistance, and mol. wt. cut-off with increasing temp. and incubation period. Thus, heating 700 mm calcium alginate beads for 20 min at 130 DegC resulted in a 23% redn. in diam., 70% in
