The Hawaiian strain (CB4856) of Caenorhabditis elegans is one of the most divergent from the canonical laboratory strain N2 and has been widely used in developmental, population, and evolutionary studies. To enhance the utility of the strain, we have gener ...

Background: In this paper, we present a framework for improving the accuracy of fixed-step methods for Monte Carlo simulation of discrete stochastic chemical kinetics. Stochasticity is ubiquitous in many areas of cell biology, for example in gene regulatio ...

Transmitter release at synapses is driven by elevated intracellular Ca2+ concentration (Ca2+) near the sites of vesicle fusion. Ca2+ signals of profoundly different amplitude and kinetics drive the phasic release component during a presynaptic ac ...

The ionotropic 5HT3 receptor was expressed in transiently transfected mammalian cells, yielding an unprecedented high concn. of up to 12 million receptors per cell. Receptor traffic in the plasma membrane of live cells was obsd. continuously over 24 h by fluorescence scanning confocal microscopy. This was possible by using 5HT3 receptor-specific fluorescent ligands with high binding affinity and low off-rate to pulse label receptors at any time after appearance on the cell surface, and label subsequently th

A review. Cell-based assays are the predominant means of assessing activation of ionotropic receptors and G protein-coupled receptors (GPCRs), but are limited by their inherent complexity. Here we present an approach to immobilize receptors stably and with defined orientation in microarrays on sensor surfaces. Using optical and elec. methods we were able to follow ligand binding, receptor activation and receptor deactivation. Our findings are a novel approach for functional screening of pharmacol. interesti

The molecular properties of fibrin-based matrices, such as fibrillar structure and covalent modifications with adhesion domains, influence the angiogenic behavior of human umbilical vein endothelial cells (HUVECs) in vitro. The fibrillar structure of fibrin-based matrices was influenced by pH but not by covalent incorporation of exogenous adhesion domains. Native fibrin-based matrices polymerized at pH 10 formed organized and longitudinally oriented fibrin fibrils, which provided a good angiogenic substrate

The ability of organisms, or individual cells to sense and to react to different external and internal stimuli (light, hormones, odorants, taste) is crucial for their survival. I will report on our recent progress to investigate mol. signal recognition and transduction processes by a combination of surface sensitive phys. techiques and mol. biol. methods. Controlled self-assembly of proteins in a functionally active form on sensor surfaces is a prerequisite for investigating mol. interactions by surface-sen

In this work the authors have shown that stable gigaohm seals over micrometer-sized holes can be obtained in a time-frame of seconds by the electrophoretic self-positioning of charged lipid membranes. For the first time, single-channel events were recorded on silicon microstructures. Self-integrating channel-forming peptides, such as alamethicin are expected to have applications in screening assays based on single-channel recordings, including high-speed DNA sequencing and metabolite detection. In the futur

The effects of acetylcholine (ACh) on excitatory postsynaptic potentials (EPSPs) evoked by stimulating Schaffer-commissural afferents and on ionophoretically applied L-glutamate ligands, were investigated in CA1 neurones of hippocampal slices using current- and voltage-clamp techniques. 2. ACh produced a transient suppression followed by a long-lasting facilitation of EPSPs. The facilitation was also seen in Cs(+)-filled cells under voltage-clamp conditions. Both suppressing and facilitating effects were

The effect of acetylcholine (ACh) on intracellular responses to ionophoretic application of N-methyl-D-aspartate (NMDA) was examined in rat hippocampal slice. Recordings were obtained from CA1 neurons under current- and voltage-clamp conditions. Drugs were applied topically by ionophoretic and microdrop techniques. ACh produced an atropine-sensitive potentiation of responses to NMDA. The effect of ACh on NMDA receptor-mediated responses was independent of changes in voltage or potassium conductances caused
