Androgen Receptor Signaling in in vivo models of Estrogen Receptor-positive Breast Cancer and Normal Breast Epithelium

Steroid hormone receptors (HRs) are ligand-activated transcription factors that play a pivotal role in breast development and carcinogenesis. It is well established that estrogen receptor (ER) signaling is a major proliferative driver in estrogen receptor ...

Nanogels with Metal‐Organic Cages as Functional Crosslinks

Kay Severin, Chaolei Hu

A dinuclear metal-organic cage with four acrylate side chains was prepared by self-assembly. Precipitation polymerization of the cage with N-isopropylacrylamide yielded a thermoresponsive nanogel. The host properties of the cage were retained within the ge ...

Role of the androgen receptor in the human breast epithelium

Fabio De Martino

Breast cancer is the most common malignancy in women worldwide and understanding the risk factors affecting it is key for prevention. The relative risk of developing breast cancer is related to exposure to both endogenous hormones and exogenous natural and ...

Socioeconomic development predicts a weaker contraceptive effect of breastfeeding

Mathias Lerch

The contraceptive effect of breastfeeding remains essential to controlling fertility in many developing regions of the world. The extent to which this negative effect of breastfeeding on ovarian activity is sensitive to ecological conditions, notably mater ...

C/EBP alpha mediates the growth inhibitory effect of progestins on breast cancer cells

Cathrin Brisken, Valentina Scabia

Steroid hormones are key gene regulators in breast cancer cells. While estrogens stimulate cell proliferation, progestins activate a single cell cycle followed by proliferation arrest. Here, we use biochemical and genome-wide approaches to show that proges ...

A high resolution LC-MS targeted method for the concomitant analysis of 11 contraceptive progestins and 4 steroids

Cathrin Brisken, Fabio De Martino, Marie Shamseddin, Csaba Ferenc László

In the context of hormonal contraception and hormone replacement therapy (HRT), many women are exposed to exogenous hormones. Current use of hormonal contraception with combined ethinyl estradiol and different progestins bestows a breast cancer relative ri ...

Sex-specific changes in gene expression in response to estrogen pollution around the onset of sex differentiation in grayling (Salmonidae)

Oliver Michele Selmoni

The synthetic 17 alpha-ethinylestradiol (EE2) is a common estrogenic pollutant that has been suspected to affect the demography of river-dwelling salmonids. One possibility is that exposure to EE2 tips the balance during initial steps of sex differentiatio ...

Progestins used for hormonal contraception in Switzerland: study of their effects on the breast epithelium

Marie Shamseddin

Breast cancer is the most commonly diagnosed cancer and the most common cause of cancer death among women. Hormones have been shown to have a key role in breast development and carcinogenesis. Repeated activation of progesterone receptor (PR) signaling as ...

Characterization of cancer stem cell subpopulations from patient-derived xenografts of advanced prostate cancer

Markus Germann

Androgen-deprivation therapy is the standard treatment for prostate cancer. Despite the initial response, a fraction of cases manifest progression to castration-resistant prostate cancer. PCa recurrence is possibly due to androgen-independent cancer stem c ...

Polyphenic trait promotes liver cancer in a model of epigenetic instability

Didier Trono, Marco Cassano, Suk Min Jang

Hepatocellular carcinoma (HCC) represents the fifth most common form of cancer worldwide and carries a high mortality rate due to lack of effective treatment. Males are eight times more likely to develop HCC than females, an effect largely driven by sex ho ...

Risk prediction of developing venous thrombosis in combined oral contraceptive users

Emmanuelle Logette, Valérie Buchillier

Background Venous thromboembolism (VTE) is a complex multifactorial disease influenced by genetic and environmental risk factors. An example for the latter is the regular use of combined oral contraceptives (CC), which increases the risk to develop VTE by ...

Identification of the Link Between the Hypothalamo-Pituitary Axis and the Testicular Orphan Nuclear Receptor NR0B2 in Adult Male Mice

Kristina Schoonjans

The small heterodimer partner (SHP, nuclear receptor subfamily 0, group B, member 2; NR0B2) is an atypical nuclear receptor known mainly for its role in bile acid homeostasis in the enterohepatic tract. We previously showed that NR0B2 controls testicular f ...

Direct noninvasive estimation of myocardial tricarboxylic acid cycle flux in vivo using hyperpolarized 13C magnetic resonance

Rolf Gruetter, Arnaud Comment, Tian Cheng, Hongxia Lei, Josefina Adriana Maria Bastiaansen

BACKGROUND: The heart relies on continuous energy production and imbalances herein impair cardiac function directly. The tricarboxylic acid (TCA) cycle is the primary means of energy generation in the healthy myocardium, but direct noninvasive quantificati ...

Intra- and interspecific social challenges modulate the levels of an androgen precursor in a seasonally territorial tropical damselfish

Studies on different vertebrate groups have provided evidence that androgen levels in males increase after competitive social interactions during the breeding season, as postulated by the Challenge Hypothesis. However, social modulation of androgen levels ...

No Scope for Social Modulation of Steroid Levels in a Year-Round Territorial Damselfish

Both latitude and mating system have been proposed to shape relationships between steroid hormone levels and social behavior. Recently it has been postulated that species with long lasting non-seasonal territorial behavior have low androgen responsiveness. ...

The Role of LEKTI in the Fate Determination of Keratinocyte Stem Cells

Alessandro Wataru Amici

SPINK5 (serine protease inhibitor Kazal-type 5) encodes the proteinase inhibitor LEKTI (lympho-epithelial Kazal-type related inhibitor). In skin, LEKTI expression is restricted to the stratum granulosum of the epidermis and the inner root sheath of hair fo ...

PAR2 absence completely rescues inflammation and ichthyosis caused by altered CAP1/Prss8 expression in mouse skin

Friedrich Beermann

Altered serine protease activity is associated with skin disorders in humans and in mice. The serine protease channel-activating protease-1 (CAP1; also termed protease serine S1 family member 8 (Prss8)) is important for epidermal homeostasis and is thus in ...

Cyclin D1 Enhances the Response to Estrogen and Progesterone by Regulating Progesterone Receptor Expression

Cathrin Brisken

Estrogen and progesterone are the defining hormones of normal female development, and both play critical roles in breast carcinogenesis. Cyclin D1 is a breast cancer oncogene whose amplification is linked to poor prognosis in estrogen and progesterone rece ...

Acquisition et recalage informatique d'images confocales de tissus denses : application à l'étude de la papille folliculaire de vibrisse de souris

Analysis of the 3D structure of a biological sample is an important step to model cell and tissue dynamics. As a model system, we set up to determine the number of nuclei in a follicular papilla using confocal imaging and 3D computer reconstruction. The fo ...

Control of thrombin signaling through PI3K is a mechanism underlying plasticity between hair follicle dermal sheath and papilla cells

Yann Barrandon

In hair follicles, dermal papilla (DP) and dermal sheath (DS) cells exhibit striking levels of plasticity, as each can regenerate both cell types. Here, we show that thrombin induces a phosphoinositide 3-kinase (PI3K)-Akt pathway-dependent acquisition of D ...
