Mechanism-driven screening of membrane-targeting and pore-forming antimicrobial peptides

The rise of antibiotic resistance has generated an urgent demand for the discovery of new antimicrobial peptides (AMPs), prompting the development of various screening strategies. However, the specific function mechanisms of AMPs are often overlooked during the screening and optimization processes. In this study, we introduce a mechanism-driven screening approach that employs machine learning-based computational models to identify peptide sequences that target membranes and form pores. This approach explici

Developement of a gateway compatible system for specific DNA integration in mouse embryonic stem cells and evaluation of the effect of nucleosome positioning on gene expression

An emerging model views gene regulation as a competition for regulatory sequences between transcription factors and nucleosomes at thermodynamic equilibrium. A computational model of the DNA affinity of nucleosomes was recently developed allowing to predict the nucleosome occupancy of DNA sequences. The aim of this project was to investigate the effect of different predicted nucleosome occupancies of an important transcription factor binding site on mammalian gene expression. To this end, a two site and thr

Transient Gene Expression in Mammalian Cells: Promises and Challenges for Medical Biotechnology

Florian Maria Wurm, Lucia Baldi Unser

Recombinant protein-based biologics are increasingly relevant in the pharmaceutical industry. Currently, mammalian cell-based bioprocesses are a routine manufacturing procedure for complex proteins such as recombinant antibodies, and the number of new biologics produced by mammalian cells is progressively increasing. In addition, a growing number of proteins need to be rapidly screened to identify new candidates for clinical trials. To help fill this need, rapid mammalian-based bioprocesses have been establ

Studies on scaffold design for tissue engineering applications : contribution of synthetic, natural and recombinant materials

The aim of tissue engineering is to regenerate tissue for the purpose of repairing or replacing diseased or injured tissue. Therefore, cells that are able to proliferate are seeded on scaffolds that provide mechanical stability and direct the three-dimensi ...

100-liter transient transfection

Florian Maria Wurm, Martin Jordan, Philippe Pierre Girard, Madiha Derouazi

This is the first report of two successful 100 l scale transienttransfections in a standard stirred bioreactor. More than half a gram of a monoclonal antibody (IgG) were produced in less than 10 days using a technology called large-scale transient gene expression(LS-TGE). Suspension adapted HEK 293 EBNA SF cells were transfectedwithin a 150 l (nominal) bioreactor by a modified calcium phosphateco-precipitation method with more than 75 mg of plasmid DNA per run.A mixture of three different plasmids, one enco

Transient gene expression: recombinant protein production with suspension-adapted HEK293-EBNA cells

Florian Maria Wurm, Horst Pick, Kirstin Friedrich

Transient gene expression (TGE) in mammalian cells at the reactor scale is becoming increasingly important for the rapid production of recombinant proteins. We improved a process for transient calcium phosphate-based transfection of HEK293-EBNA cells in a 1-3 L bioreactor volume. Cells were adapted to suspension culture using a commercially available medium (BioWhittaker, Walkersville, MD). Process parameters were optimized using a plasmid reporter vector encoding the enhanced green fluorescent protein (EGF

alpha- and beta-homogalactonojirimycins (alpha- and beta-homogalactostatins): Synthesis and further biological evaluation

Pierre Vogel

The homoiminosugars alpha- and beta -homogalactonojirimycins were prepared from a common intermediate, tetra-O-benzyl-D-galacto-heptenitol 6, by way of highly stereoselective reaction sequences involving, as the key steps, an internal amidomercuralion (a-epimer) and a double reductive amination (beta -epimer). alpha -Homogalactonojirimycin retains a large part of the potent activity of the parent galactonojirimycin and 1-deoxygalactonojirimycin as an inhibitor of alpha -galaclosidases. However, by contrast

Lentivirus gene transfer in murine hematopoietic progenitor cells is compromised by a delay in proviral integration and results in transduction mosaicism and heterogeneous gene expression in progeny cells

Human immunodeficiency virus type 1-based lentivirus vectors containing the green fluorescent protein (GFP) gene were used to transduce murine Lin(-) c-kit(+) Sca1(+) primitive hematopoietic progenitor cells. Following transduction, the cells were plated into hematopoietic progenitor cell assays in methylcellulose and the colonies were scored for GFP positivity. After incubation for 20 h, lentivirus vectors transduced 27.3% +/- 6.7% of the colonies derived from unstimulated target cells, but transduction wa

Transfecting mammalian cells: optimization of critical parameters affecting calcium-phosphate precipitate formation

Florian Maria Wurm, Martin Jordan

DNA-calcium phosphate co-precipitates arise spontaneously in supersaturated solutions. Highly effective precipitates for transfection purposes, however, can be generated only in a very narrow range of physico-chemical conditions that control the initiation and growth of precipitate complexes. The concentrations of calcium and phosphate are the main factors influencing characteristics of the precipitate complex, but other parameters, such as temperature, DNA concentration and reaction time are important as w

Expression of Herpes simplex virus type 1 glycoprotein C antigens in Escherichia coli

Florian Maria Wurm

DNA fragments encoding structural information of the Herpes simplex virus type 1 (HSV-1) glycoprotein C (gC) gene were cloned into pUC plasmids [Vieira and Messing, Gene 19 (1982) 259-268]. None of the hybrid plasmids were able to direct the synthesis of significant amounts of gC related peptides. Several of the plasmid-bearing strains, however, exhibited inhibition characteristics which can be correlated with the presence on the plasmid of specific gC gene sequences. After insertion of gC DNA fragments int
