Dynamics of antibodies from cryo-electron tomography

Résumé

The issue of protein dynamics and its implications in the biological function of proteins are arousing greater and greater interest in molecular biology. In cryo-electron tomography experiments one takes several snapshots of a given biological macromolecule. In principle, a large enough collection of snapshots may then be used to calculate its equilibrium configuration in terms of the experimentally accessible degrees of freedom, and hence estimate its potential energy. Consequently, one could analyze the biological functions of biomolecules by directly accessing their dynamics.

Microcrystal electron diffraction

Microcrystal electron diffraction, or MicroED, is a CryoEM method that was developed by the Gonen laboratory in late 2013 at the Janelia Research Campus of the Howard Hughes Medical Institute. MicroED is a form of electron crystallography where thin 3D crystals are used for structure determination by electron diffraction. Prior to this demonstration, macromolecular (protein) electron crystallography was only used on 2D crystals, for example.

Protein dynamics

Proteins are generally thought to adopt unique structures determined by their amino acid sequences. However, proteins are not strictly static objects, but rather populate ensembles of (sometimes similar) conformations. Transitions between these states occur on a variety of length scales (tenths of Å to nm) and time scales (ns to s), and have been linked to functionally relevant phenomena such as allosteric signaling and enzyme catalysis.

Structure des protéines

La structure des protéines est la composition en acides aminés et la conformation en trois dimensions des protéines. Elle décrit la position relative des différents atomes qui composent une protéine donnée. Les protéines sont des macromolécules de la cellule, dont elles constituent la « boîte à outils », lui permettant de digérer sa nourriture, produire son énergie, de fabriquer ses constituants, de se déplacer, etc. Elles se composent d'un enchaînement linéaire d'acides aminés liés par des liaisons peptidiques.
