Résumé

Anthrax toxin-receptor interactions are critical for toxin delivery to the host cell cytoplasm. This review summarizes what is known about the molecular details of the protective antigen (PA) toxin subunit interaction with either the ANTXR1 and ANTXR2 cellular receptors, and how receptor-type can dictate the low pH threshold of PA pore formation. The roles played by cellular factors in regulating the endocytosis of toxin-receptor complexes is also discussed.

Anthrax toxin

Anthrax toxin is a three-protein exotoxin secreted by virulent strains of the bacterium, Bacillus anthracis—the causative agent of anthrax. The toxin was first discovered by Harry Smith in 1954. Anthrax toxin is composed of a cell-binding protein, known as protective antigen (PA), and two enzyme components, called edema factor (EF) and lethal factor (LF). These three protein components act together to impart their physiological effects. Assembled complexes containing the toxin components are endocytosed.

Pore-forming toxin

Pore-forming proteins (PFTs, also known as pore-forming toxins) are usually produced by bacteria, and include a number of protein exotoxins but may also be produced by other organisms such as apple snails that produce perivitellin-2 or earthworms, who produce lysenin. They are frequently cytotoxic (i.e., they kill cells), as they create unregulated pores in the membrane of targeted cells. PFTs can be divided into two categories, depending on the alpha-helical or beta-barrel architecture of their transmembrane channel that can consist either of Alpha-pore-forming toxins e.

En biochimie, un récepteur est une protéine de la membrane cellulaire ou du cytoplasme ou du noyau cellulaire qui se lie spécifiquement à un facteur spécifique (un ligand, tels un neurotransmetteur, une hormone, une molécule médicamenteuse, une toxine, un ion calcium, ou une protéine spécifique de la membrane d'un virus...), induisant une réponse cellulaire à ce ligand. Les modifications du comportement du récepteur protéique induites par le ligand conduisent à des modifications physiologiques qui constituent les « effets biologiques » du ligand.
