From Brittle to Flexible: Influence of Glucose‐Based Additives on Cellulose Nanocrystal Self‐Assembly in Suspension and Dried Photonic Films

Cellulose nanocrystal (CNC) films can exhibit reflective and iridescent coloration due to their self‐assembled structure, but are brittle. This study compares glucose (monosaccharide), maltose (disaccharide), and gluconic acid (acidic sugar) as plasticizers and links suspension self‐assembly behavior to the optical and mechanical properties of the resulting films. Glucose‐plasticized films have been previously studied, but maltose and gluconic acid have not yet been applied in this capacity. Here, glucose a

Unveiling functional-metabolic synergy in the healthy brain: multivariate integration of dynamic [ 18 F]FDG-PET and resting-state fMRI

Introduction Despite accounting for only 2% of body weight, the human brain requires significant amounts of glucose, even at rest, underscoring the importance of functional-metabolic relationships. Previous studies revealed moderate associations between resting-state fMRI functional connectivity (FC) and local metabolism via [ 18 F]FDG-PET, yet much remains to be understood, particularly regarding their coupling between functional and metabolic networks. Methods To this end, we employed multivariate Partial

Position-specific carbon isotope analysis of glucose and tree-ring cellulose by Orbitrap mass spectrometry

The isotopic compositions of environmentally relevant compounds like glucose and cellulose carry traces of the metabolic processes they have undergone. However, many of the relevant fractionations are imparted at specific atomic positions, whereas most traditional isotope analyses measure the average isotopic composition of the whole molecule. Here, we present the first method for position-specific isotope analysis (PSIA) of glucose and cellulose via Electrospray-Ionization (ESI)-Orbitrap mass spectrometry.

Covalent conjugation of glucose oxidase on single-walled carbon nanotubes for glucose sensing

Glucose sensing and monitoring are crucial for biological and medical applications. Compared to existing methods, real-time detection and long-term monitoring are still required. Single-walled carbon nanotubes (SWCNTs) have excellent optical properties for sensing applications, which provide the possibility for designing a new generation of glucose sensors. In this study, we describe a method for covalently conjugate glucose oxidase (GOx) on SWCNTs as an optical glucose sensor. The functional groups are int

Toward Quantitative Measurements of Enzyme Kinetics by Dissolution Dynamic Nuclear Polarization

Geoffrey Bodenhausen, Sami Jannin, Aurélien Bornet, Jonas Milani

Dissolution dynamic nuclear polarization (D-DNP) experiments enabled us to study the kinetics of the enzymatic phosphorylation reaction of glucose to form glucose-6-phosphate (G6P) by hexokinase (HK), with or without the presence of an excess of G6P, which ...

Increasing the sensitivity of enzyme-based biosensors using copper-chloride and EDTA

Nico de Rooij

The effect of CuCl2 and EDTA on increasing the sensitivity of enzyme biosensors was evaluated. The experiments revealed that successive exposure of these solutions to our biosensors can increase the sensitivity of the glucose and lactate biosensors up to 88% and 50%, respectively.

Sirtuins as regulators of metabolism and healthspan

Since the beginning of the century, the mammalian sirtuin protein family (comprising SIRT1-SIRT7) has received much attention for its regulatory role, mainly in metabolism and ageing. Sirtuins act in different cellular compartments: they deacetylate histones and several transcriptional regulators in the nucleus, but also specific proteins in other cellular compartments, such as in the cytoplasm and in mitochondria. As a consequence, sirtuins regulate fat and glucose metabolism in response to physiological c

Cell metabolism monitoring using a microfluidic cartridge with enzyme-based microreactors

Nico de Rooij

We present a novel microfluidic approach for on-line analysis of cell metabolism by simultaneous measurement of two important indicators (glucose and lactate) in a small volume (100 µl) of cell-culture medium. The in-vitro study of cell metabolism is especially valuable for evaluating the effect of toxins on physiochemical state of living organisms

Measurements of Glycogen Metabolism in the Living Brain

To maintain brain function, the combustion of glucose for the generation of energy (ATP) is essential, yet the brain maintains fuel storage in the form of glial glycogen. Although brain glycogen levels are lower than those in the liver, they exceed the normal free glucose concentration in brain several-fold. Under normoxic conditions, when a partial glucose supply deficit is present, brain glycogen can provide fuel for extended periods of time. Brain glycogen metabolism is active, but comparatively slow, co

New Technologies for Nanobiosensing and their Applications to Real-Time Monitoring

Amperometric biosensors are complex systems and they require a combination of technologies for their development. The aim of the present work is to exploit existing technologies for new purposes in order to develop nanostructured biosensors for the real-time detection of multiple metabolites in cell culture flasks. The fabrication of five Au working electrodes onto silicon substrate is achieved with CMOS compatible microtechnology. Each working electrode presents an area of 0.25 mm2, so structura

Amyloid-beta aggregates cause alterations of astrocytic metabolic phenotype: impact on neuronal viability

Pierre Magistretti

Amyloid-beta (Abeta) peptides play a key role in the pathogenesis of Alzheimer's disease and exert various toxic effects on neurons; however, relatively little is known about their influence on glial cells. Astrocytes play a pivotal role in brain homeostasis, contributing to the regulation of local energy metabolism and oxidative stress defense, two aspects of importance for neuronal viability and function. In the present study, we explored the effects of Abeta peptides on glucose metabolism in cultured ast

A novel enzyme entrapment in SU-8 microfabricated films for glucose micro-biosensors

Nico de Rooij

The present work investigates the utilisation of the widely used SU-8 photoresist as an immobilisation matrix for glucose oxidase (GOx) for the development of glucose micro-biosensors. The strong advantage of the proposed approach is the simultaneous enzyme entrapment during the microfabrication process within a single step, which is of high importance for the simplification of the BioMEMS procedures. Successful encapsulation of the enzyme GOx in "customised" SU-8 microfabricated structures was achieved thr
