Ether lipid remodeling during neuronal differentiation prevents ferroptosis

Ferroptosis is a form of cell death driven by iron-dependent lipid peroxidation, with specific lipid species playing key roles in modulating susceptibility. Among these, ether lipids have shown conflicting effects, being linked to both protection and sensitization. Here, we dissect the relationship between lipid structure and ferroptosis sensitivity and explain how ether lipids exert context-dependent effects. Ether lipids can promote ferroptosis through a metabolic bias towards the accumulation of polyunsa

Single-walled carbon nanotube biosensor for real-time monitoring of nitric oxide in inflammatory responses

Osteoarthritis (OA) is a degenerative inflammatory joint disease affecting millions of people worldwide. The early detection of OA and the continuous monitoring of its progression are essential for managing the disease. In this study, we develop an optical system for monitoring OA-related inflammation by detecting nitric oxide (NO), a molecule that is overproduced in joints during OA. The NO sensor is based on fluorescent single-walled carbon nanotubes (SWCNTs) coated with single-stranded DNA (ssDNA). The s

Preclinical Mechanistic Evaluation of Hyaluronan/Niacinamide (Vitamin B3) Hydrogels: Toward an Enhanced Viscosupplement System with Ancillary Anti-Arthritic Attributes

Osteoarthritis (OA), a degenerative joint disease primarily affecting the hips and knees, is characterized by multifactorial dysregulation of chondrocyte homeostasis and currently lacks curative treatment options. Intra-articular hyaluronic acid (HA) injections have clinically provided symptomatic relief for three decades; however, HA’s rapid in vivo degradation by free radicals and hyaluronidases limits its efficacy. We hypothesized that adding niacinamide (vitamin B3) to linear HA hydrogels would provide

Quantitative imaging of lipid transport in mammalian cells

Eukaryotic cells produce over 1,000 different lipid species that tune organelle membrane properties, control signalling and store energy1,2. How lipid species are selectively sorted between organelles to maintain specific membrane identities is largely unclear, owing to the difficulty of imaging lipid transport in cells3. Here we measured the retrograde transport and metabolism of individual lipid species in mammalian cells using time-resolved fluorescence imaging of bifunctional lipid probes in combination

Oxytocin release modulates acute neuroinflammation and improves brain development after pediatric traumatic brain injury

Pediatric traumatic brain injury (TBI) is a leading cause of death and disability early in life in infants, and its neurodevelopmental consequences cannot currently be effectively treated. Since TBI is associated with neuroinflammation, modulation of the post-injury neuroinflammatory response is a promising strategy. Oxytocin is thought to have anti-inflammatory properties and appears to play a role in clinical interventions that improve brain development in neonates. However, the underlying mechanisms rema

Peroxisome Staining in Mammalian Cells Using Peroxisome-Specific Probes

Peroxisomes are ubiquitous organelles with essential roles in human health, and their dysfunction leads to a spectrum of genetic disorders. Visualization of peroxisomes is key to identifying peroxisome dysfunction; however, progress has been limited by the lack of peroxisome-specific probes. By developing PeroxiSPY, peroxisome-specific probes that mimic natural peroxisome metabolites, we enabled real-time detection, tracking, and quantification of peroxisome dynamics in live mammalian cells. Additionally, t

New Insights into the LANCL2-ABA Binding Mode towards the Evaluation of New LANCL Agonists

The lanthionine synthetase C-like (LANCL) proteins include LANCL2, which is expressed in the central nervous system (CNS) and in peripheral tissues. LANCL2 exhibits glutathionylation activity and is involved in the neutralization of reactive electrophiles. Several studies explored LANCL2 activation as a validated pharmacological target for diabetes and inflammatory bowel disease. In this context, LANCL2 was found to bind the natural product abscisic acid (ABA), whose pre-clinical effectiveness in different

A benthic foraminifera species respond to anoxia with a strong metabolic shift suggesting a state of dormancy

Low-oxygenation events are more and more frequent and strong on continental shelves and in coastal areas where renewal of bottom waters is slow. Among the meiofauna living in such environments, foraminifera are among the most tolerant to the lack of oxygen. Some benthic foraminiferal species are able to survive hypoxia, and even anoxia, for weeks to months. Different species must have developed different mechanisms for survival - hypotheses include reduction of the metabolism, symbiosis with bacteria, or de

Kinetic modeling of the yeast sphingolipid metabolism identifies prevalent mutant strains through integration of lipidomic data profiles

Sphingolipids are abundant components of eukaryotic cells. Their localization in the plasma membrane allows for the cell to carry out multiple important functions for its viability. One of the most important roles of the sphingolipids is their ability to form, along with other lipid compounds, complex structures which attribute to the membrane of the cell its functional role to selectively allow transport of molecules, participate in signaling, react under heat stress and regulate growth. In yeast multiple
