Small, nonpolar cyclic peptides can both bind challenging targets and cross cell membranes, making them attractive for addressing currently undruggable targets such as many protein–protein interactions (PPIs). However, developing such compounds de novo without prior information about lead structures such as natural ligands or fragments remains a notable challenge. Here we show that functional screening of structurally highly diverse cyclic peptide libraries synthesized at nanomole scale allows identificatio

The electrostatic potential of lipid membranes is an important metric to gain insight into the physiological state of cells. Second harmonic (SH) imaging has emerged as a powerful tool to perform in-situ and in-vivo electrostatic potential measurements, either using targeted probes, or label-free by using water as a probe. In this work we compare both modalities, by performing SH imaging on freestanding lipid membranes (FLMs) in aqueous solutions. Correlating the SH intensity to an applied external potentia

Membrane fusion is ubiquitous in myriad biological processes, including presynaptic neurotransmitter release, myoblast fusion, virus entry, and fertilization. It is often considered in the presence of fusogenic proteins, which mediate the fusion by pulling the membranes into contact to overcome the high energy barrier imposed by hydration repulsion. Here we uncover an alternative pathway: membrane fusion arising from electroporation induced by transmembrane potentials in absence of proteins. Using biased mo

Ferroptosis is a form of cell death driven by iron-dependent lipid peroxidation, with specific lipid species playing key roles in modulating susceptibility. Among these, ether lipids have shown conflicting effects, being linked to both protection and sensitization. Here, we dissect the relationship between lipid structure and ferroptosis sensitivity and explain how ether lipids exert context-dependent effects. Ether lipids can promote ferroptosis through a metabolic bias towards the accumulation of polyunsa

Eukaryotic cells produce over 1,000 different lipid species that tune organelle membrane properties, control signalling and store energy1,2. How lipid species are selectively sorted between organelles to maintain specific membrane identities is largely unclear, owing to the difficulty of imaging lipid transport in cells3. Here we measured the retrograde transport and metabolism of individual lipid species in mammalian cells using time-resolved fluorescence imaging of bifunctional lipid probes in combination

Amphiphilic lipid formulations, such as self-emulsifying drug delivery systems, offer advantages for enhancing drug release control and expanding their applicability across various administration routes. By integrating microfabrication techniques with these lipid-based systems, additional functionalities such as controlled drug release can be introduced. This can broaden lipid's potential for advanced biomedical and pharmaceutical applications. However, lipids face major fabrication challenges due to their

Tension propagates in lipid bilayers over hundreds of microns within milliseconds, seemingly precluding the formation of tension gradients. Nevertheless, plasma membrane tension gradients have been reported in migrating cells and along growing axons. Here, we show that the mechanosensitive, fluorescent membrane probe Flipper-TR visualizes membrane tension gradients in artificial and cellular membranes. Images of tension gradients allow their quantitative characterization, showing that they are long-ranged a

Lipidomics, a subfield of metabolomics, is an emerging field where hundreds to thousands of lipid species are simultaneously identified. Given the ubiquity and diverse biological roles of lipids, lipidomics offers valuable insights into mechanisms and the discovery of biomarkers related to environmental stressors that affect the cellular physiology and their numerous biochemical pathways. The major source of lipids in the atmosphere are the biogenic particles (bioaerosols) e.g., bacteria, fungi, pollen, pla

The development of fluorescence-based molecular imaging has revolutionized cell biology allowing the visualization of specific biomolecules at the microscopic and, more recently, at the nanoscopic scale while in their relevant biological contexts. Nonetheless, despite the imaging toolkit for biologists interested in exploring the subcellular localization and dynamics of proteins and nucleic acids has expanded exponentially in the last decades, the means to visualize and track lipids in cells did not develop

Biological membranes are highly dynamic and complex lipid bilayers, responsible for the fate of living cells. To achieve this function, the hydrating environment is crucial. However, membrane imaging typically neglects water, focusing on the insertion of p ...

We consider the numerical approximation of lipid biomembranes, including red blood cells, described through the Canham-Helfrich model, according to which the shape minimizes the bending energy under area and volume constraints. Energy minimization is performed via L2- gradient flow of the Canham-Helfrich energy using two Lagrange multipliers to weakly enforce the constraints. This yields a highly nonlinear, high order, time dependent geometric Partial Differential Equation (PDE). We represent the biomembran

Lipids are a large and diverse group of biological molecules with roles in membrane formation, energy storage and signaling. Cellular lipidomes may contain tens of thousands of structures, a staggering degree of complexity whose significance is not yet fully understood. High-throughput mass spectrometry-based platforms provide a means to study this complexity, but the interpretation of lipidomic data and its integration with prior knowledge of lipid biology suffers from a lack of appropriate tools to manage
