Significance
Cerebral autoregulation (CA) reflects the dynamic coupling among cerebral blood flow (CBF), intracranial pressure (ICP), and arterial blood pressure (ABP); its failure contributes to secondary brain injury. Existing bedside methods rely on indirect or spatially limited CBF surrogates and cannot resolve microvascular flow dynamics across space, depth, and time. Aim
To develop, optimize, and apply a scalable, noncontact time-resolved laser speckle contrast imaging (TR-LSCI) platform

Background: Chronic anxiety is common, disabling, and often refractory to current therapies. Mounting evidence implicates mitochondrial abnormalities in anxiety-related phenotypes. Urolithin A (UA), a gut microbiota-derived metabolite known to enhance mito ...

Early identification of drug toxicity is essential yet challenging in drug development. At the preclinical stage, toxicity is assessed with histopathological examination of tissue sections from animal models to detect morphological lesions. To complement this analysis, toxicogenomics is increasingly employed to understand the mechanism of action of the compound and ultimately identify lesion-specific safety biomarkers for which in vitro assays can be designed. However, existing works that aim to identify mo

Background The pathological process of Alzheimer's disease (AD) typically takes decades from onset to clinical symptoms. Early brain changes in AD include MRI-measurable features such as altered functional connectivity (FC) and white matter degeneration. The ability of these features to discriminate between subjects without a diagnosis, or their prognostic value, is however not established. Methods The main trigger mechanism of AD is still debated, although impaired brain glucose metabolism is taking an inc

Reward signals encoded in the mesolimbic dopaminergic system guide approach/seeking behaviors to all varieties of life-supporting stimuli (rewards). Differences in dopamine (DA) levels have been found between dominant and submissive animals. However, it is ...

Purpose To probe the cardiac metabolism of carbohydrates and short chain fatty acids simultaneously in vivo following the injection of a hyperpolarized 13C-labeled substrate mixture prepared using photo-induced nonpersistent radicals. Methods Droplets of m ...

D-amino acid oxidase (DAO) is an enzyme that catalyzes the degradation of D-amino acids in the body. Here, we explored the possibility of detecting D-amino acid oxidase activity by monitoring its metabolism in the rat kidney after a bolus injection of hyperpolarized D-[1-13C]alanine. Our data show that D-alanine is readily converted to lactate only when the DAO enzyme is not inhibited, indicating that the observed metabolism is that of DAO.

Exposure to early adversity is implicated in the development of aggressive behaviour later in life in some but not all individuals. The reasons for the variability in response to such experiences are not clear but may relate to pre‐existing individual diff ...

Parkinson's disease motor symptoms are treated with levodopa, but long‐term treatment leads to disabling dyskinesia. Altered synaptic transmission and maladaptive plasticity of corticostriatal glutamatergic projections play a critical role in the pathophys ...

Aspartic acid is involved in several central metabolic pathways, including gluconeogenesis, the urea cycle, de novo nucleotide synthesis, the malate-aspartate shuttle, and via transmination the TCA cycle. The conversion in vivo of hyperpolarized [1-13C]pyruvate to aspartate, via pyruvate carboxylation to oxaloacetate, has been reported in rat kidney [1] and in the mouse liver [2]. Additionally, the metabolism of hyperpolarized [1-13C]aspartate to oxaloacetate has been noted in cultured cells supplemented wi

The microbiome has been demonstrated to play an integral role in the maintenance of many aspects of health that are also associated with aging. In order to identify areas of potential exploration and intervention, we simultaneously characterized age-relate ...

Ex vivo and in vivo studies on liver metabolism using hyperpolarized [1-13C]pyruvate report do not agree on whether hyperpolarized bicarbonate metabolite production results from pyruvate oxidation or gluconeogenesis. This study tested the ability of hyperpolarized [1-13C]pyruvate to probe gluconeogenesis in the liver of intact rats. While conversion to hyperpolarized bicarbonate was detected in the liver of fasted rats, treatment with the phosphoenolpyruvate carboxykinase inhibitor 3-mercaptopicolinc acid r

Peripheral nerves are often interfaced with cuff-based and microchannel-based nerve implants so that neural activity may be stimulated, inhibited and/or recorded. In vivo evaluation of these interfaces is mandatory but often time-consuming, and focused on single implant design. In vitro studies can provide further design freedom to the bioelectronic engineer, but neuron cultures do not replicate accurately in vivo, in-nerve conditions. In order to overcome these restrictions, we developed a microchannel ele

Lesions to the anterior thalamic nuclei (ATN) in rats produce robust spatial memory deficits that reflect their influence as part of an extended hippocampal system. Recovery of spatial working memory after ATN lesions was examined using a 30-day administra ...

Although cocaine abuse is up to three times more frequent among schizophrenic patients, it remains unclear why this should be the case and whether sex influences this relationship. Using a maternal immune activation model of schizophrenia, we tested whethe ...
