This lecture introduces the Hi-C technique, starting from the cross-linking and fragmentation of chromatin to the mapping of DNA fragments that form loop structures. By isolating and sequencing these fragments, researchers can analyze the spatial interactions between distal genomic regions, reconstructing the three-dimensional conformation of chromatin.
